Important Distinction: Weight Loss vs Body Composition

Most peptides with metabolic research do not directly cause significant weight loss in the way GLP-1 agonists (semaglutide, tirzepatide) do. What some peptides do is improve body composition — reducing fat mass, preserving or increasing lean muscle mass, and improving metabolic markers. These are related but distinct outcomes.

MOTS-c: The Exercise Mimetic

MOTS-c’s most compelling metabolic research shows improved insulin sensitivity and resistance to diet-induced obesity in animal models. The 2021 Nature Aging study’s finding that it rises in response to exercise positions it as a potential metabolic signal rather than a direct fat-loss agent.

GH Secretagogues and Body Composition

GH has established effects on body composition — reducing fat mass and preserving lean mass. GH secretagogues that raise IGF-1 toward the upper-normal range can have meaningful body composition effects over 3-6 month protocols. This is not dramatic weight loss but can be clinically meaningful for patients with age-related sarcopenia or metabolic decline.
