The Distinction

Thymosin Beta-4 (Tฮฒ4) is a naturally occurring 43-amino-acid protein present in virtually all human cells. TB-500 is a synthetic peptide corresponding to amino acids 17-23 of Thymosin Beta-4 โ€” specifically the actin-binding domain. It is a 7-amino-acid fragment, not the full protein.

Researchers identified the actin-binding domain as the portion of Thymosin Beta-4 most responsible for its observed biological activity. By synthesizing just this fragment, researchers can study the mechanism more precisely, and pharmaceutical manufacturers can produce a more consistent, synthesizable compound than the full 43-amino-acid protein.

Does the Fragment Have the Same Effects?

This is the key scientific question. Research suggests the actin-binding fragment retains most of the relevant biological activity โ€” the effects on cell migration, wound healing, and tissue repair observed in full Thymosin Beta-4 research appear to be substantially mediated through this domain. But the full protein may have additional activities not captured by the fragment.
